中文摘要: 实现生物样品的低剂量三维成像,对于基础生命科学研究和医学应用具有重要意义。图像重建技术是X射线层析成像的核心和关键,本项目拟采用等斜率迭代这一新算法进行图像重建。该方法是基于伪极傅里叶变换,用等斜率方式取代传统CT的等角度方式采集投影,利用少量投影获得高精度图像,从而降低样品辐射剂量。本项目拟充分利用上海光源,通过对线站设备的适当改进,优化和应用等斜率迭代算法,开展生物样品的低剂量、高分辨、定量化三维成像方法研究。针对上海光源线站的特点,将等斜率迭代算法分别和扫描透射X射线成像与相衬成像结合,实现在投影数目较少或数据不全情况下生物样品的三维高分辨成像;并探索活体生物的三维成像。希望通过本项目研究,丰富和发展上海光源线站成像方法,提升我国大科学装置在活体生物成像及三维高分辨成像方面的研究能力。

英文摘要: Low-dose three-dimensional imaging of biological samples is of great significance for life sciences and medical applications. Because image reconstruction technique is the key of X-ray tomography, a new algorithm – equally sloped tomography will be used for image reconstruction in the project. This algorithm is based on the pseudo-polar fast Fourier transform. In order to reduce radiation dose, a small amount of projections, which are acquired at equally-sloped angles, is used to obtain high-precision image of samples. We will make use of Shanghai Synchrotron Radiation Facility and make equipment improvements at appropriate beamlines, to carry out low dose, high-resolution, quantitative three-dimensional imaging of biological samples. In combination with phase contrast imaging and scanning transmission X-ray imaging, equally sloped tomography can provide three-dimensional imaging of biological samples in the case of a relatively small number of projections or incomplete data. We will also explore imaging of living organisms. We hope this work can not only open a door to low dose three-dimensiona imaging of biological samples, but also improve the research capabilities in imaging at Shanghai Synchrotron Radiation Facility.
