中文摘要: 间歇性爆发性障碍是一种以反复出现的发作性暴力攻击冲动为特征的精神障碍,此类患者常常带来许多经济损失和法律问题,已日益成为司法精神病学的关注热点。有推测环境与遗传因素(特别是DNA甲基化)在其发生中起了协同作用,但具体机制仍有待阐明。 本研究拟以间歇性爆发性障碍人群为研究对象,采用纵向对照研究方法,探索暴力性间歇性爆发性障碍患者与普通人群在愤怒情绪水平变化、外周细胞(淋巴细胞和口腔粘膜细胞)中愤怒情绪调控相关基因(5-HT和DA系统)的基因多态性与DNA甲基化水平,以及外显暴力攻击行为上的差异,并分析三者之间的动态变化关联。以此探讨间歇性爆发性障碍的发病中DNA甲基化通过影响愤怒情绪调控从而调控暴力攻击行为的可能机制,为暴力攻击行为的预防和干预提供新的思路;并考察淋巴细胞与口腔粘膜细胞作为外周标记在精神疾病的DNA甲基化调控相关研究中的生物学意义,为其他心理行为障碍的表观遗传学研究提供理论依

英文摘要: Intermittent explosive disorder (IED)is a mental disorder characterized by recurrent episodes of serious aggressive outbursts, which cause a lot of economic damage and legal issues, and become the focus point of forensic psychiatry. It is hypothesized that the Interactive between environmental and genetic factors ( especially DNA methylation ) plays an key issues in the formation of IED, but the exact mechanism remains to be elucidated. A longitudinal study will be carried out to explore the change of state-trait anger and DNA methylation ( 5-HT and DA) in eripheral cells ( lymphocytes and buccal mucosa cells ) among intermittent explosive disorder patients, in order to explore the role of state-trait anger and DNA methylation in the regulation of violence behavior in these patients. Through this research, we hope to provide new ideas to the prevention and intervention of violence behavior; besides, the value of lymphocytes and buccal mucosa cells as a biological markers in the research of DNA methylation in mental disorders will also be disscussde, thus to privide theoretical basis for epigenetics research in other psychological and behavioral disorders.
