中文摘要: 生殖与内分泌是甲壳类重要的研究内容。甲壳类红色素聚集激素(RPCH)通过刺激GSH与MF两种生殖激素的分泌从而促进卵巢发育,是一种重要的神经肽。受激素分泌检测技术的限制,迄今RPCH调控作用的研究尚停留在个体与器官水平上。拟穴青蟹是我国养殖产量最高的海水蟹类,是重要的水产养殖种类。本项目拟开展RPCH对青蟹卵巢发育的调控机制及应用基础研究,内容包括:(1)RPCH结构与特性,从分子和细胞水平查清RPCH的结构、分布与表达特性;(2)RPCH对分泌活动的调控,从单个细胞水平揭示RPCH对离子通道电流与胞吐分泌的影响;(3)RPCH生殖调控技术,根据RPCH诱导卵巢成熟的作用,综合青蟹发育阶段与培育环境,建立一种基于RPCH的新型促熟技术。本研究将从细胞和分子水平揭示RPCH的调控机制,同时建立一种新型促熟技术,对深化甲壳类内分泌学研究和促进养殖发展均具有重要意义。

英文摘要: Reproduction and endocrine are important research issues in crustaceans. Red pigment concentrating hormone (RPCH), an important neuropeptide, accelerates ovarian development by stimulating the secretion of gonad-stimulating hormone (GSH) and methyl farnesoate (MF). However, limited by the measurement technique of hormones, the study on the regulation of RPCH still remains at the level of individual and organ. The mud crab, Scylla paramamosain, is a commercially important aquatic species, with the highest yield in Chinese marine crabs. This program is intended to undertake the research on the mechanism and application of RPCH in Scylla paramamosain. The contents include 1) the structure and characteristic of RPCH, clarifying the structure, distribution and expression of RPCH at molecular and cellular level; 2) the role of RPCH on endocrine, revealing the effect of RPCH on the ionic channel currents and exocytosis at cellular level; 3) the reproductive control technique of RPCH, establishing a new technique of inducing ovarian maturation based on the effect of RPCH on the ovarian stages and culture environment. This research will identify the regulation mechanism of RPCH at the molecular and cellular level, and establish a new technique of inducing ovarian maturation.
