中文摘要: 多取代烯丙醇(胺)的不对称异构化反应是合成β-位有手性的羰基化合物的重要方法,但是由于双键上的取代基位阻效应的影响导致该类反应很难进行,目前还没有有效解决该类问题的方法。本项目计划对多取代烯丙醇(胺)异构化的反应机理进行深入的研究,从中找出影响的关键因素,尝试通过改变配体而调控催化剂,或在反应体系中加入适当的添加剂(路易斯酸,胺)和使用弱配位反应溶剂,或在底物上引用一些导向基团,从而实现一系列多取代烯丙基类化合物的不对称异构化反应,实现一些重要的药物中间体和天然化合物的合成。同时,对简单及多取代烯丙基类化合物的动力学拆分进行优化,高效地得到光学纯的目标分子,并将该方法应用到一些重要手性中间体的合成。

英文摘要: Asymmetric isomerization of allylic alcohols (amines) is a straightforward and efficient reaction to afford optically active carbonyl compounds. However, the substrate often has a limitation and catalytic activity tends to decrease as the degree of substitution of the olefinic bond increases. As a result, there is no efficient and general method to realize these asymmetric isomerization reactions. We expected to find out critical factors effecting these reactions by investigating their reaction mechanisms.Tuning the electronic and steric factors of the ligands to modify the catalysts, Using proper additives (Lewis acids or amines) and solvents with different coordinating ability, and introducing some coordinating groups on double bond of allyl compouds will be the methods employed to achieve these asymmetric isomerization reactions.Kinetic resolution of allyl compounds with multiple substituations will also be studied.These synthetic methods developed will find their use in synthesizing important chiral building blocks for pharmaceutical chemistry and natural products.
