中文摘要: 水稻株型改良一直被认为是一个提高粮食产量的有效途径,当前我国超级稻育种就是以"株型形态与生理机能相结合,理想株型和杂种优势相结合"为指导思想,因此对水稻株型调控基因的克隆和功能研究将有助了解水稻株型形成的分子机制。本研究利用9311 EMS诱变获得了一个抗倒直立株型隐性突变体ept1,通过其与日本晴的回交BC2F2群体,已经将该基因定位在了195kb的区间内,我们将继续利用构建好的BC3F2大群体,开展EPT1的精细定位,并最终克隆该基因。同时拟通过构建不同的功能和表达载体及与其它株型控制基因构建双突变,对EPT1进行功能研究。并利用EMS回复诱变寻找抑制基因,同时开展基因芯片、酵母双杂交对上下游基因和互作蛋白进行筛选、验证和分析。初步探明EPT1基因在水稻植株形态建成的遗传机制及分子调控机理,为分子设计育种,创建水稻抗倒伏新种质打下基础。

英文摘要: Rice plant type improvement has been considered to be an effective way to increasing food production. plant type combining with physiological function and ideal plant with heterosis is the guiding ideology in china super rice breeding. So the cloning and functional studies of rice plant type genes will facilitate to understand the molecular mechanism of rice plant type development. In this study, the 9311 lodging resistance recessive mutant ept1 with the erect plant type had been obtained from EMS treated. By the backcross BC2F2 population with Nipponbare, the gene was final mapped to an interval of 195kb. We will furter fine mapping and cloning the gene with a larger BC3F2 population. The EPT1 functional studies will be carry out by constructing different functions and expression vector and through the double mutant analyzing with other plant type gene. We will also start EMS recovery mutagenesis, gene chip and yeast two-hybrid system to finding, verifying and anlyzing of these suppressor gene, downstream gene and interacting proteins of EPT1, which will preliminarily ascertain the genetic and molecular mechanism of the EPT1 gene, provide theoretical basis for molecular design breeding and germplasm innovation of lodging resistance..
