中文摘要: 合成阴、阳离子均可与蛋白质活性基团形成氢键作用的离子液体,研究皮胶原在非水介质-离子液体中的溶解行为和在超临界CO2流体中的脱脂行为,获得在温和物理条件下胶原的高效提取方法;针对传统的先解构降解后功能改性造成胶原固有结构遭到破坏的技术缺陷,提出对皮胶原进行先结构修饰和功能改性,后进行生化解构、降解的技术路线,获得在不破坏胶原三股螺旋结构前提下其目标结构和性能的控制方法;在此基础上,进一步研究胶原的重要结构参数(如三股螺旋结构、聚集态结构、分子量及其分布)对胶原溶液性能(如可溶性、热稳定性、凝胶性等)及生物学性能(如抗原性、凝血性、生物相容性、可吸收性等)的影响,获得胶原结构-性能的内在构-效规律。展开皮胶原高效提取方法、解构方法、结构修饰及功能改性方法及其构-效关系等关键科学问题研究,对高效利用生皮胶原具有十分重要地意义。

英文摘要: A type of ionic liquid which its anionic and cationic can combine with the active groups of collagen by hydrogen bonds are synthesised and the dissolving and degreasing behavior of skin collagen in ionic liguid and supercitical CO2 fluid respectivity are studied so as to achieve a high efficient way to extract collgen under mild condition;By means of structure and function moditication fristly then bio-chemical dicomposition, a controlling method for desired structure and property without destroy the triple-helix structure can be achieved.On this base, the relationship between typical structure factors(such as triple helix structure, aggregation state, molecular weight and distribution) and solution characteristics (i.e, soluble property, thermal stability and gel-forming property) as well as bio charateristics(such as antigenicity,blood coagulation,sbaorbability, biocompatibility) will be investigated in detail.It is believed that to investigate the extraction method,dicomposition method, structure and function modification methods togather with the structure-activity of collagen is very usefull and significant for high effficiency useage of collagen.
